A novel mutation (Arg629Trp) in the androgen receptor hinge region is found to impair N/C domain interaction in the presence of normal androgen receptor binding and nuclear localization, and to cause severe undermasculinization at birth and resistance to androgens, thus confirming a unique regulatory role for this region in androgen receptor function.

Orlistat accelerates gastric emptying and markedly reduces the postprandial secretion of the anorexigenic hormones glucagon-like peptide-1, peptide YY, and cholecystokinin. However, these alterations in gastrointestinal hormone concentrations may increase appetite and food consumption, and should be considered when using lipase inhibitors for the treatment of morbid obesity.

Mild short-term experimental hyperthyroidism increases whole body protein turnover and breakdown before any measurable changes in energy expenditure, glucose, and fat metabolism, suggesting that amino acid and protein metabolism is an early and primary target for thyroid hormone action in humans.

The transcription factor 7-like 2 (TCF7L2) rs7903146 T-allele associates with hepatic insulin resistance and diminishes glucose-stimulated plasma insulin secretion in Danish twins. There is no evidence of a role of TCF7L2 gene expression in the regulation of glucose homeostasis in subcutaneous fat or muscle.

Fetal growth restriction induces changes in body composition and metabolism suggestive of a higher insulin sensitivity independently from birth weight itself, reflecting adaptive changes to an adverse fetal environment, thus leading to understanding the association between low birth weight and the risk of type 2 diabetes later in life.

Despite a linear decrease in GH secretion and peak stimulated GH levels with increasing body mass index in healthy overweight and obese women, IGF-I levels are not commensurately reduced. Androgens may contribute to this relative preservation of IGF-I secretion despite reduced GH secretion.

Serum TSH secretion profiles in five thyrotropinoma patients are described. As in other pituitary tumors, secretion was characterized by increased pulsatility, basal release, reduced pattern regularity, and phase shifting of the diurnal rhythm. These features imply altered properties of the thyrotrope and (partial) loss of cell-cell communication.

A loop of interleukin (IL)-1{alpha} expression by resident stromal cells amplifies the paracrine induction of stromal matrix metalloproteinase-1 by epithelial-derived IL-1{alpha} in the degradation of endometrium during menstruation and dysfunctional bleeding. Ovarian steroids control the release of epithelial IL-1{alpha} and the synthesis of stromal IL-1{alpha}, respectively.

The Wnt/{beta}-catenin pathway is activated in primary pigmented nodular adrenocortical diseases and adrenocortical adenomas with PRKAR1A mutations, suggesting a cross-talk between the cAMP and the Wnt/{beta}-catenin pathways in adrenocortical tumor development. In addition, the occurrence of an additional hit of a CTNNB1 somatic mutation is associated with larger or more aggressive adrenocortical tumors.

In malignant thyroid nodules, low sodium-iodide symporter (NIS) mRNA expression was associated with strong intracellular NIS protein staining, probably relating to the NIS protein's inability to migrate to the plasma membrane and to the low iodide uptake seen in malignant tumor cells.
